If you haven't uploaded/installed it yet, configure b2config.php and upload it to the '/' folder.
If you have installed it from before;
a) take a backup of your database tables (just in case)
b) configure b2config.php to point to the '/' folder instead of /previoususedfolder/
c) check your .css and images :p
d) move the files (can be done in ws ftp if you allow it), OR _download_ all the files to your HDD from the /previousfolder/, upload it to /.

If you've CHMOD'ed the relevant pages (like index.php/b2.php, b2comments.php or b2commentspopup.php) 766, you can log on go to Template and alter them there (by doing direct HTML editing).
If you want to build a template from scratch, which I recommend, go ahead and check out this short tutorial on what you need first and foremost to get the thing working:

You should also edit / replace b2layout.css or make your own internal/external css file. I recommend external for easier updating and smaller php files.
If you don't have HTML/CSS experience but want to learn, check out:
http://www.w3schools.com
If you don't have HTML/CSS experience and doesn't want to learn, hire a webdesigner or use a WYSIWYG* editor (Like Frontpage, DreamWeaver etc.)..
